I am a single mom of 4 kids and because of this would like to set up a support group for moms in South Africa. On joining they would then be entitled to certain incentives at various shops eg, car maintenance, grocer, hairdresser and the list goes on. There would also be a legal, medical and informative centre where they could get help when needed.
Must I charge the moms a fee for joining? How do I get the businesses to join? The 24/7 care line how can I make money from this?

Sounds like a great idea. You may have trouble getting people to pay to join a support group though. I htink it would be good if you could look into getting a sponsor (use "In association with" or "Sponsored by" in any of your communications)who could help out with the financial support to get you started.
You could approach local businesses to supportyou to - car maintenance etc (as you mention below) and they too could be assoicated with you. Good exposure and credibility for them and financial help from you.
You need to do a business plan (there's one on this site I've seen in the article section), so your clear of your objectives. You can then approach local businesses (or larger ones) for help.
24/7 care line - again you look look at your service provider supporting you.
Are you looking to set this up alone, or do you have any one else to partner up with?

I agree with Wyatt's comments. You could also approach local schools and colleges to see if any other women want to go into business with you. Do you have any contacts in the legal, medical etc worlds? They could obviously help you out too.

I wouldn't charge single mums (who maybe struggling financially). You should look at ways to provide help and support for free
